Purification and some physical properties of phenylalanine hydroxylase from rat liver.

چکیده
Phenylalanine hydroxylase has been obtained from rat liver in a form that is 85 to 95% pure. The enzyme exists as two electrophoretically distinguishable isozymes, each of which is capable of existing as a monomer (51,000 to 55,000 molecular weight), dimer (llO,OOO), and tetramer (210,000). The dimers of each isozyme will dissociate to the monomeric forms by dilution of the enzyme or by increasing the temperature from 0 to 30”.

Purification and state of activation of rat kidney phenylalanine hydroxylase.
Phenylalanine hydroxylase, the enzyme that catalyzes the irreversible hydroxylation of phenylalanine to tyrosine, was purified from rat kidney with the use of phenyl-Sepharose, DEAE-Sephacel, and gel permeation high pressure liquid chromatography. Our most highly purified fractions had a specific activity in the presence of 6-methyltetrahydropterin, of 1.5 mumol of tyrosine formed/min/mg of pro...
متن کاملThe isolation and properties of phenylalanine hydroxylase from human liver.
Phenylalanine hydroxylase was prepared from human foetal liver and purified 800-fold; it appeared to be essentially pure. The phenylalanine hydroxylase activity of the liver was confined to a single protein of mol.wt. approx. 108000, but omission of a preliminary filtration step resulted in partial conversion into a second enzymically active protein of mol.wt. approx. 250000. Human adult and fu...
متن کاملMechanism of phenylalanine regulation of phenylalanine hydroxylase.
The mechanism of phenylalanine regulation of rat liver phenylalanine hydroxylase was studied. We show that phenylalanine "activates" phenylalanine hydroxylase, converting it from an inactive to active form, by binding at a true allosteric regulatory site. One phenylalanine molecule binds per enzyme subunit; it remains at this site during catalytic turnover and, while there, cannot be hydroxylat...
متن کاملPurification and characterization of phenylalanine hydroxylase-stimulating protein from rat liver.
Phenylalanine hydroxylase-stimulating protein markedly stimulates phenylalanine hydroxylase activity under various conditions and thus may serve a critical function in phenylalanine metabolism. At pH values above neutrality, or in the presence of phospholipids such as lysolecithin, when the naturally occurring cofactor, tetrahydrobiopterin, is used, the specific activity of phenylalanine hydrox...
متن کاملProperties of Phenylalanine Hydroxylase of Cultured Hepatoma
The kinetic and immunologic properties of phenylalanine hydroxylase of adult rat liver were compared to the properties of the similar enzyme present in cultured H4-II-E-C3 hepatoma cells. The enzymes from the two sources could not be distinguished by the K, values for either phenylalanine or 6,7-dimethyltetrahydropterin. Analysis by double immunodiffusion showed that phenylalanine hydroxylase f...
